Chamberlain served as the 32nd Governor of Maine, holding office from 1867-1871. He was admitted to Bowdoin College in 1848, and after his graduation in 1852, he entered the Bangor Theological Seminary. In the fall of 1855, he returned to Bowdoin where he became an instructor in logic and natural theology. In addition to Latin and German, Chamberlain mastered French, Arabic, Hebrew and Syriac.

Chamberlain volunteered for the Union Army during the Civil War, serving in the 20th Maine Volunteer Infantry Regiment. He fought at the Battle of Fredericksburg, and earned renown for his gallantry at the Battle of Gettysburg, for which he was awarded the Medal of Honor. In 1865, he fought at the Battle of Five Forks, and was given the honor of commanding the Union troops at the surrender ceremony for the infantry of Robert E. Lee's Army at Appomattox Court House in Virginia. After the war, he entered politics, ultimately being elected Governor of Maine.  He served one term, and after leaving office, returned to Bowdoin College to serve as its President until 1883.
